DESCRIPTION:Webinar summary: \nLymphocytic choriomeningitis virus (LCMV) is a single-stranded RNA arenavirus commonly used in laboratory animal models to examine acute and chronic viral infections and immunopathology. However\, there is wide variation in LCMV biosafety practices between institutions\, based on differing interpretations of guidelines and risk data. This lack of consensus can pose a challenge to the biosafety professional who may be evaluating LCMV use at their facility. The first part of this presentation will compare the two most common laboratory strains of LCMV\, demonstrate the variation in biosafety policy between institutions\, and discuss one institution’s recent approach to navigating the use of this virus. \nOutside of the laboratory\, LCMV has also become a promising viral vector with potential use in immunizations and immunotherapy. The second portion of this presentation will examine the applications of LCMV in a clinical setting\, discussing the biosafety practices necessary for the safe handling of LCMV-based human gene therapies\, focusing on the clinical risk assessment associated with recombinant forms of LCMV. SUMMARY:BSL-3 Operations\, Maintenance & Verification Training
DESCRIPTION:This introductory course is a four-day training designed to certify personnel for safe entry into an operational BSL-3 laboratory. This course consists of a mixture of classroom and practical training that includes an introduction to microbiology\, biosafety\, risk assessment concept\, BSL-3 lab characteristics\, emergency management and personal protective equipment (PPE) use including procedures for donning and doffing PPE with N95 or various PAPR respirators. \nThis course is suitable for O&M personnel and other non-research staff who may need to enter BSL-3 facilities to perform maintenance or make repairs. The ANSI/ASSE Z9.14 Standard focuses on performance verification of engineering controls specifically related to the ventilation system features of BSL-3/ABSL-3 facilities. Z9.14 is the only guidance document that provides a methodology to verify ventilation systems in such facilities. The standard provides a risk-based approach to reaching containment goals appropriate to the risk of the agent and the laboratory activity. This course will provide a complete overview of the standard as well as provide practical hands-on training on how to assemble the team\, perform the test methodology\, and determine the suitability of example BSL-3/ABSL-3 facilities based on risk assessment. \nHeld at our National BSL-3 Training Lab with full testing functionality and equipment\, this course is suitable for BSL-3 laboratory managers\, biosafety professionals\, facilities managers\, architects\, engineers and commissioning agents.

Selection and Use of Personal Protective Equipment (PPE)\nRecent infectious disease outbreaks highlighted the need for the proper use of Personal Protective Equipment (PPE). The guidance on use of appropriate PPE varies greatly and can often be contradicting and therefore confusing to the end user. Furthermore\, availability of different PPE often varies depending on the situation and location. Selection of appropriate PPE should be based on a comprehensive risk assessment that takes in consideration the different hazards present\, as well as the procedures/activities to be conducted. Often times\, locations where PPE is needed vary as well\, from a well-controlled laboratory environment to collection room or a field location\, and even in a patient room setting. Each of these situations require staff to be able to perform a comprehensive risk assessment\, often in a fast-evolving situation and with a minimal understanding of specifications associated with different PPE components available to them. This course will provide overview of different PPE options available; specifications associated with each option\, and address a risk assessment process for selection of appropriate PPE for different environments. The examples will include some of the most common issues and considerations when selecting PPE such as supply chain issues as well as strategies to address behavioral concerns that may arise. In groups\, participants will develop risk assessment for different scenarios\, present their risk assessment to the class\, and evaluate each other’s PPE selection based on their risk assessment. Instructors will guide participants through the process by providing critical feedback on the PPE features and selection process as they progress. The course will go through an effective risk assessment process which is critical for proper selection of appropriate PPE. Keeping it Going: Maintaining and Improving a Select Agent Program over the Long Term\nKeeping a select agent program going can be difficult\, especially in the face of ever-changing regulatory requirements and limited resources. Established procedures may suddenly become unacceptable\, interrupting research\, and frustrating laboratorians. Likewise\, a single unexpected adverse event can put an entire program at risk. Being prepared to deal with such changes and events is critical to maintaining a robust program. Anticipating future challenges can be even more advantageous\, elevating a program from good to great. A proactive approach can minimize the impact of new requirements and reduce duration and frequency of crises sparked by sudden\, unexpected requirements or events. This course will explore strategies for maintaining and improving an existing program\, including how to anticipate and respond to new requirements. The focus will be on U.S. select agent requirements\, although a small section on comparable requirements in the international community will be included. Strategies will be based on the instructors’ experience with their program with additional input solicited from class participants during open discussions. Topics will include select agent program history; effective oversight; efficiently meeting ongoing requirements; reporting\, responding to\, and analyzing incidents; suitability program case studies; inactivation requirements\, including “failure” investigation; and inspection preparation and response. DESCRIPTION:Empirical Research in Biosafety: Filling Key Data Gaps\nPresented By: Rocco Casagrande\, Ph.D.\,\nExecutive Chairman\, Gryphon Scientific\nIn this session\, we will present data from our empirical work in biosafety and our work exploiting existing data sets to inform biosafety. We will discuss our methodological framework for studying aerosols generated by laboratory accidents\, and present information on the aerosols produced by dropping microtiter plates and tissue culture flasks. Also in the physical sciences\, we will present data on the rate that conical centrifuge tubes leak and the frequency that splashes occur when opening microcentrifuge tubes via various opening methods. We will discuss the rate of spills and splashes when pipetting as drawn from experiments using volunteers and blinded samples in clinical laboratories. Interestingly\, this experiment also sheds light on the ability of the researcher to know when they are making mistakes and take corrective action. We will present data on the rate at which needle sticks can be expected in the laboratory. We will examine how biosafety findings are distributed amongst laboratories in several institutions and what can be learned about the culture of biosafety. We will discuss how knowledge of the frequency and causes of accidents can lead to means to improve reproducibility in the life sciences.
